Chronic exposure to glucocorticoids during critical neurodevelopmental periods leads to lasting shifts in neuronal type distribution and overall brain architecture

Ilknur Safak Demirel, Pia Giraudet, Malgorzata Grochowicz, Anthi C. Krontira, Leander Dony, Tim Schäfer, Elisabeth Binder, Cristiana Cruceanu · FENS Forum 2024

The brain undergoes important growth and plasticity during prenatal development. Altered activation of the glucocorticoid receptor (GR) system is one of the factors mediating stress effects during this time, likely through transcriptional dysregulation. To investigate these processes in a human-specific in vitro system, we used induced pluripotent stem cell-derived 3-dimensional brain organoids. To determine cell-type specific GR activation response, we profiled the transcriptomes of thousands of individual cells using single-cell transcriptomic analyses following glucocorticoids exposure acut

Transcriptomic and sublaminar dynamics of the developing human frontal cortex: A novel approach

Janja Kopić, Parthiv Haldipur, Leonarda Grandverger, Jure Krasić, Zeljka Krsnik · FENS Forum 2024

Our recent study showed specific molecular and cellular differences in sublaminar dynamics of the dorsal/dorsolateral and orbitobasal part of the prospective human frontal cortex (Kopić et al, 2023). More particularly, during early midfetal development, dorsal part of the prospective frontal cortex showed waves of migratory cells, representing future deep projection neurons (DPN) characterized by diverse molecular markers, while orbitofrontal cortex displays "double" cortical plate pattern shown by DPN markers (TBR1, TLE4, SOX5). Cell-sparse layer in between these “two plates” shows DCX+ cells
