Control of RVM to spinal cord pain descending pathway by vlPAG-SST neurons in a physiopathological context

Juliette Viellard, Amalia Lambert-Ringuet, Frédéric Naudet, Rabia Bouali-Benazzouz, Abdelhamid Benazzouz, Pascal Fossat · FENS Forum 2024

Pain is an adaptive and primordial aspect of mammal’s physiology whose role is to warn against potential harmful situations and prevent aggravation of actual physical damages. However, many patients suffer from chronic pain, which overpasses pain’s adaptive trait becoming pathological. Nociceptive signal transmission is first relayed by the spinal cord before reaching the brain through ascending pathways. The control of spinal integration descending pathway is orchestrated by the periaqueductal gray (PAG) and rostral ventromedial medulla (RVM). In the RVM, ON, OFF and neutral cells, are respec

Sex differences in nociceptor regeneration after burn injury

Chiara Nappi, Espe Selva, Francisco J. Taberner · FENS Forum 2024

Burns of diverse etiologies pose a significant health burden, resulting in varying levels of tissue damage and pain. Despite extensive research utilizing mouse models to elucidate burn-induced pain and recovery, discrepancies across different methodologies underscore the intricate complexity of burn pain and the necessity of standardized burn injury paradigms. To address this limitation, we developed the Burn Injury by Repeated Heat Exposure (BIRHE) model. By laying the hind paw for 10-seconds to a 52°C plate for 10 or 20 repetitions we reproducibly induce distinct pain intensities. We used th

The effect of diabetes on myelin homeostasis

Melvin Alappat, Hanns Ulrich Zeilhofer, Sevasti Gaspari · FENS Forum 2024

Peripheral diabetic neuropathy (DN) affects 50% of Diabetes Mellitus (DM) patients and approximately 40-50% progress to painful diabetic neuropathy (pDN). Risk factors for pDN include high alcohol consumption, obesity, and low vitamin D levels, all associated with impaired oligodendrocyte progenitor cell (OPC) differentiation. Additionally, studies indicate that depletion of mature myelinating oligodendrocytes induces nociceptive responses in mice. These findings suggest that DM might disrupt oligodendrocyte dynamics, causing defects in central nervous system (CNS) myelin and the onset of neur
