Differential neuroinflammatory responses to immune challenges in male and female rats prenatally exposed to alcohol

Carolina Luft, Kingston Wong, Victoria Vella, Parker Holman, Tamara Bodnar, Charlis Raineki · FENS Forum 2024

Prenatal alcohol exposure (PAE) leads to immune system dysfunction and increased susceptibility to autoimmune diseases. To explore potential mechanisms by which PAE induces neuroimmune dysfunction, the current study examined the impact of PAE on toll-like receptor 4 (TLR4) and TLR7/8 activation. Pregnant rats were divided into two groups: PAE - liquid ethanol diet ad libitum, and CON - pelleted control diet ad libitum. Male and female offspring were challenged in adulthood with LPS (40 µg/kg; activates TLR4), R848 (1 mg/kg; activates TLR7/8), or DMSO (vehicle), followed by brain sample collect

Neuronal network dysfunction and neurodegeneration mediated by TLR7/8-activated microglia depend on the immunological context

Lennart Söder, Andrea Lewen, Amr Elgez, Babak Khodaie, Oliver Kann · FENS Forum 2024

Innate Toll-like receptors (TLRs) play a critical role in activating microglia in various diseases of the CNS. Our study aimed to examine the widely unknown consequences of stimulation of TLR7/8, which are known to recognize single-stranded RNA (ssRNA), e.g. from viruses. For this, we conducted electrophysiological local field potential recordings of neuronal gamma-oscillations (30-70 Hz), biochemical assays, and immunofluorescence in rat organotypic hippocampal slice cultures. Our findings reveal that single exposure (48 h) to the TLR7/8 agonist Resiquimod (R848) induces the release of pro-in
