Abstract

Aims: Urocortin-1 (UCN1) is a corticotropin-releasing hormone (CRH)-like neuropeptide associated traditionally with stress, anxiety and depression. This study aimed to investigate the behavioral effects of the UCN1 fragments on anxiety-like and depression-like behavior.
Methods: For this purpose, C57BL/6 mice were injected intracerebroventricularly (ICV) with UCN1 and its fragments, UCN1(1-20) or UCN1(21-40). After 30 min, the behavior of mice was examined in an elevated plus-maze test (EPMT) and a forced swim test (FST). When a significant effect was observed, CRH antagonists, such as antalarmin (selective for CRH1 receptor) and astressin2B (selective for CRH2 receptor) were also injected ICV 30 min before the administration of agonists.
Results: While UCN1 and UCN1(1-20) produced anxiogenic and depressant effects, UCN1(21-40) induced anxiolytic and antidepressant behavior in mice. The effects of UCN1 and UCN1(1-20) were abolished by antalarmin, but not astressin2B, and those of UCN1(21-40) were inhibited by CRH2, but not CRH1 antagonist.
Conclusions: These results suggest a functional polarity within the UCN1 molecule regarding its impact on anxiety and depression, which can be mediated by two distinct, CRH1 and CRH2 receptors, a preclinical observation that may have clinical significance.
