GLIAL ACTIVATION ALONG THE VISUAL PATHWAY IN AN ANIMAL MODEL OF GLAUCOMA

Methods: Adult C57/BL6 male and female mice were submitted to unilateral OHT in the left eye (OHT eye) by laser photocoagulation, while the contralateral right eye (CONTRA eye) was not manipulated and remained normotensive. Intraocular pressure (IOP) was measured at different time points until sacrifice, at 15 and 28 days after the laser-induced surgery. Brains were extracted and processed for the posterior analysis of microglial and macroglial cell markers, by immunohistochemical techniques using Iba-1 and GFAP antibodies, respectively, in the dorsolateral geniculate nucleus (dLGN), superior colliculus (SC), and visual cortex (V1).
Results: Following OHT on the left eye we observed an activation of glial cells on the contralateral right hemisphere, which accounts for 95% of the projections from the retina. Similar results were observed at the two time points analysed, for both Iba1 and GFAP expression, although the observed effects were more robust at day 28.
Conclusions: Present findings provide evidence for glial activation in the main relay visual pathway nuclei of the contralateral hemisphere to the eye in which glaucoma was induced; however, the physiological significance of these changes remain to be clarified.
