Microglia were isolated from mixed glial cultures and added to 7-day-old tri-cultures. Upon isolation, microglia were kept in presence of supplements (M-CSF, TGF-β and cholesterol) for microglial support in serum-free environments. The quadruple culture was monitored for 14 days using live-cell imaging and immunocytochemistry. To evaluate immune response cultures were exposed to LPS. Microglial numbers peaked at day 7, coinciding with less cell death. While integration of microglia did not impact neuronal or astrocytic development, it may have stimulated the differentiation of oligodendrocytes.
In summary, we developed a quadruple culture model incorporating microglia that serves as a promising tool for screening nutritional compounds aimed at reducing neuroinflammation.
