Abstract

The knowledge of the molecular and cellular mechanisms that are involved in brain development is rather limited. Moreover, while many extracellular cues and specific receptors have been shown to play a role in different aspects of nervous system development, including neuron migration and axon guidance, examples of intracellular signaling effectors involved in these processes are sparse. In the present work, we show that the atypical RhoGTPase, RhoE/Rnd3, plays an important role for axon guidance and cortex development during the formation of the brain. We show that RhoE is necessary for the formation of some of the main axon tracts in the brain, such as the thalmocortical and striatal axons and that RhoE is required for proper radial migration of the cortical neurons during development. RhoE-dependent migration requires fosforylation of the protein and proper membrane localitzacioni by farnesylation. Finally, we show that RhoE interacts with FLRT3 and both cooperate in the formation of brain cortical folds.
