Recent pre-clinical studies suggest a trans-synaptic propagation mechanism of pathological proteins. However, direct investigation in humans has been hindered by limited access to high-quality samples, imaging constraints, and tissue preservation challenges.
Here, we combine tissue clearing and expansion microscopy to map AD pathology across scales in human brain tissue, from entire brain regions to the nanoscale organization of synapses. This integrated approach offers access to region- and stage-specific ultrastructural changes in pathological proteins, and to their trans-synaptic spread within mesoscale-identified disease-affected brain regions.
By advancing the histological characterization of AD pathology in human tissues, this method supports human-based research and opens new perspectives for translational research in AD.
