Abstract

The generation of diverse inhibitory neuron subtypes is critical for the formation of tegmental nuclei that regulate mood, motivation and movement. Using single-cell RNA sequencing, we identified distinct GABAergic neuron subtypes in the developing brainstem, including previously uncharacterized neuropeptidergic populations. We validated these populations and determined their anatomical distribution using multiplex RNAscope in situ hybridization. Our results refine the molecular and spatial landscape of GABAergic neuron diversity in the anterior brainstem and provide new insight into the developmental origins of inhibitory neuronal heterogeneity relevant to brainstem circuit function and behavior.
