EAE was induced in mice subjected to one of four dietary conditions: sunflower seeds, wheat kernels, a combination of both, or no food enrichment. Clinical disease severity and body weight were assessed longitudinally. Immune cell composition in the periphery and central nervous system was analyzed by flow cytometry. For mechanistic understanding, plasma lipidomic profiling and gut microbiome analyses were performed.
Initial data demonstrated that combined food enrichment significantly exacerbated EAE severity and increased disease-associated weight loss. In an expanded study, disease progression, immune profiles, and metabolic outcomes were assessed across dietary interventions to delineate the contribution of individual food components. Immunophenotyping encompassed major peripheral and CNS immune populations, while plasma lipidomics included analyses of lipid class composition and fatty acid balance. Together, these data explore potential links between dietary fat composition, immune regulation, and neuroinflammatory disease severity.
These findings indicate that high-fat food enrichment can aggravate autoimmune neuroinflammation in EAE. This work highlights the importance of dietary control in preclinical MS models and underscores nutrition as a relevant modulator of disease severity with translational implications.
