Photopharmacology in the spotlight: Spatially selective adenosinergic modulation of hippocampal excitability in the pursuit of therapeutic innovation in temporal lobe epilepsy

Marijke Vergaelen, Jeroen Spanoghe, Jeroen Missinne, Serge Van Calenbergh, Kristl Vonck, Paul Boon, Robrecht Raedt · FENS Forum 2024

The adenosine A1 receptor (A1R) is a promising therapeutic target in epilepsy by mediating neuronal inhibition. In this study, we investigated whether A1R signaling is still functional in the intrahippocampal kainic acid (IHKA) mouse model for temporal lobe epilepsy (TLE). Moreover, we evaluated the feasibility of spatial selective inhibition of specific hippocampal subregions through photopharmacology. Electrically evoked field postsynaptic potentials (fPSPs) were recorded in the CA1 and DG of acute hippocampal slices. The effect of adding 40 nM of the A1R agonist N6cyclopentyladenosine (CPA)

Role of adenosine A1 receptors in oligodendrocyte regeneration

Qilin Guo, Ting Zhang, Qing Liu, Anja Scheller, Frank Kirchhoff, Wenhui Huang · FENS Forum 2024

Adenosine A1 receptor (A1AR) signaling exerts important functions in the central nervous system (CNS). Among all CNS cell types, oligodendrocyte (OL) lineage cells possess the highest A1AR expression levels. However, in vivo data regarding the functions of A1ARs in OL lineage cells remain elusive. Here, we first generated NG2-CreERT2xA1ARfl/fl conditional knockout (cKO) mice to specifically delete A1ARs from oligodendrocyte precursor cells (OPCs) upon tamoxifen administration. Immunohistochemical analysis for OL lineage markers PDGFRα, APC CC1, as well as BrdU incorporation detected unaltered
