Cannabidiol (CBD) is a phytocannabinoid with potential as a therapy for a variety of diseases. CBD may act via cannabinoid receptors but also via other G-protein-coupled receptors (GPCRs), including the adenosine A2A receptor. Homogenous binding and signaling assays in Chinese hamster ovary (CHO) cells expressing the human version of the A2A receptor were performed to address the effect of CBD on receptor functionality. CBD was not able to compete for the binding of a SCH 442416 derivative labeled with a red emitting fluorescent probe that is a selective antagonist that binds to the orthosteri

The endocannabinoid system (ECS) has gained attention as a potential target to treat Alzheimer’s disease (AD) due to its neuroprotective, antioxidant, and anti-inflammatory properties. Consequently, natural cannabinoids like delta9-tetrahydrocannabinol (THC) and cannabidiol (CBD) have emerged as potential drugs against AD. Previous findings from our research group demonstrated that a combination of non-psychoactive doses of THC and CBD mitigates cognitive decline in an AD murine model, the APP/PS1 mice. Cognitive decline in AD is linked to the dysregulation of glutamate transmission within the

Neuronal Kv7 (KCNQ) voltage-gated potassium channels contribute to various neurophysiological functions (such as hearing, learning, and pain modulation), and, when mutated, cause distinct pathologies (such as hearing loss, epilepsy, and intellectual disability). Cannabidiol (CBD) is a pan-Kv7 modulator, Kv7.2/3 M-channel activator and EMA-approved antiepileptic. Yet, the potential therapeutic efficacy of similar phytocannabinoids on Kv7 channels remains to be studied. Here, we aim to investigate the pharmacology of cannabinoids structurally related to CBD, such as Δ9‑Tetrahydrocannabinol (THC)

Approximately 2-5% of pregnant women may use cannabidiol (CBD) on a weekly basis: an estimated 75,000 to 200,000 fetuses annually in the US alone are exposed to CBD. Identifying the impact of CBD on the developing CNS is a public health priority. CBD crosses the placenta and concentrate in breast milk. To investigate the long-term behavioral consequences of fetal CBD exposure, we compared the behavior of adult offspring from dams exposed daily to a low dose of CBD (3mg/kg s.c., E5-E18). The adult offspring was observed using Live Mouse Tracker, a methodology, based on deep learning, eliminatin

Neonatal hypoxia-ischemia is a severe condition that can result in death and/or lifelong central nervous system sequelae. Benefits of cannabidiol (CBD) result from neuroprotection asssumed to be mainly due to targeting a variety of neuronal receptors. The results of this article first show that CBD protects both primary neurons and primary glia against death induced by glucose and oxygen deprivation. Second, the expression of glial markers in an animal model of neonatal hypoxia-ischemia treated with vehicle or CBD shows that the cannabinoid was able to skew microglia towards a neuroprotective

The endocannabinoid system influences virtually all central nervous system processes: learning, memory, appetite, emotions, and more. Growing evidence suggest that modulation of the endocannabinoid system by endogenous or exogenous cannabinoids may be significant in the treatment of various diseases. Tetrahydrocannabinol (THC) is the main psychoactive substance found in cannabis has been used as a pain medication due to its sedative effect. However, a whole series of other phytocannabinoids have been isolated, such as cannabinol (CBN), cannabidiol (CBD), tetrahydrocannabivarin (THCV) and Δ9-Te
