The developmental effects of repeated antenatal dexamethasone treatment on ADP-mediated and adenosinergic signaling system in the auditory brainstem of C57BL/6 mice

Dunja Dimitrijević, Irena Lavrnja, Marija Adžić-Bukvić, Milorad Dragić, Anđela Stekić, Katarina Mihajlović, Ivan Milenković, Danijela Laketa · FENS Forum 2024

Pregnant women at risk of premature labor receive a single treatment with the synthetic glucocorticoid dexamethasone (DEX) between the 24th and 34th week of pregnancy to ensure adequate maturation of the fetal organs. Due to the postponement of the due date, several doses are often administered. Because purinergic signaling plays an important role in the development of the auditory brainstem, particularly the cochlear nucleus, we investigated whether repeated DEX treatments affect components of the ADP-mediated (NTPDase2 and P2Y1R) and adenosinergic (CD73 and A1R) signaling systems. To compare

Dual inhibition of ecto-5'-nucleotidase (CD73) and adenosine A2A receptor reduces neuroinflammation and oxidative stress in TNF, IL-1α, C1q-induced neurotoxic astrocytes

Katarina Mihajlović, Marija Adžić Bukvić, Ivana Stevanović, Milorad Dragić, Nadežda Nedeljković · FENS Forum 2024

Ecto-5'-nucleotidase (CD73) is an ecto-enzyme that hydrolyzes adenosine monophosphate to adenosine, which acts through one of four subtypes of adenosine receptors. Increased CD73 activity and expression in chronic neurodegenerative diseases leads to a higher concentration of adenosine in the extracellular space. This results in excessive activation of adenosine A2AR, which triggers proinflammatory signaling pathways that further promote neurodegeneration. Therefore, dual CD73/A2AR inhibition is a potential pharmacologic target in chronic neurodegeneration. In the present study, we used a novel

Modulating the astrocyte reactivity by blocking P2X7R and Panx1 in vitro – intercepting the neuroinflammation chronicity development

Marija Adzic Bukvic, Ivana Stevanovic, Katarina Milicevic, Nadezda Nedeljkovic · FENS Forum 2024

Neural cells and tissue injury lead to abundant release of ATP from the cell. Additionally, ATP could be cut loose through the activated Pannexin 1 hemichannels. Extracellular ATP could activate P2X7R, P2 ligand-gated ion channels, activating the inflammasome in microglia and astrocytes. P2X7R and Panx1 could create forward feedback loop, that potentiates and drives the chronicity of previously initiated neuroinflammation. Both P2X7R and Panx1 are upregulated in multiple neurodegenerative diseases, hypoxia, injuries, depression, epilepsy, and pain. Targeting one or both P2X7R and Panx1 has bec

Olfactory dysfunction as a common denominator in multiple sclerosis and Parkinson’s disease – Evidence from animal models

Andjela Stekic, Milorad Dragic, Milica Zeljkovic Jovanovic, Nadezda Nedeljkovic · FENS Forum 2024

Olfactory dysfunction is an early symptom of neurodegenerative diseases, including Parkinson’s disease (PD) and multiple sclerosis (MS). Recent data showed that 30-40% MS patients and 90 % PD patients experience olfactory dysfunction. Studies have shown that adenosine signaling has a pivotal role in olfactory processing; thus, deciphering the changes in adenosine signaling may be crucial for understanding the disease pathogenesis. Aim of this study was to characterize olfactory dysfunction from the aspect of adenosine metabolism, focusing on ecto-5’ nucleotidase (CD73) in the olfactory bulb (O

Progesterone alleviates neurological deficit and striatal ectonucleotidase activities after cerebral hypoperfusion in rats

Progesterone (P4) has shown promising results in diminishing the effects of cerebral hypoperfusion (CH) in prefrontal cortex. Therefore, our goal was to further analyze the neuroprotective mechanism of P4 in adult Wistar rats following bilateral occlusion of common carotid arteries (2VO). We investigated its impact on neurological function and activity of striatal extracellular purine metabolizing enzymes, CD39 and CD73. Following either sham or 2VO operation, for 7 consecutive days, animals (n=12) received s.c. treatments of commercial flax oil (V, 1 mg/kg) (Sham+V and 2VO+V groups) or 1.7 mg
