Benzodiazepine tolerance is a well-documented consequence of their chronic use, the mechanism of which is not yet fully understood. To investigate tolerance development in mice, we have treated C57BL/6J male and female mice with 10 mg/kg diazepam once daily for 27 days (study 1) or twice daily for 14 days (study 2). In study 1, the Spontaneous Locomotor Activity (SLA) test captured sedation consistent with the reduction in the total distance travelled by diazepam-treated mice of both genders across time, compared to vehicle-treated mice (two-way ANOVA). These effects did not diminish over time

The open field test (OFT) is widely used to score anxiety and depression levels in rodents. In the OFT, fruit flies show wall-following behavior (WAFO) similar to rodents. However, it is questionable to which extent insects can perceive emotions such as fear or depression. To characterise the influence of emotion-like states on the OFT behavior in flies, we measured both WAFO and total walking (TOWA) in the OFT in wild-type flies paired with negative or positive stimuli, altered internal states, or pharmaceutical treatment.CantonS wildtype flies treated with negatively valenced biological stre

Neuroactive steroids such as allopregnanolone that act as a positive allosteric modulator of synaptic and extrasynaptic δ-subunit-containing GABAA receptors, has shown antidepressant effects on depressive symptoms in both clinical and preclinical studies. However, the neuronal dynamics associated with these effects are still unknown. In the present study, we discovered that allopregnanolone, but not the benzodiazepine diazepam, has antidepressant-like effects in the social interaction test (SIT) of social defeat stress (SDS) model mice by decreasing intervals of repetitive social interaction (

It is not unusual to tacitly conflate conjectured inhibitory processes in cognition with inhibitory neural mechanisms. Here, we investigate whether resolution of memory interference, which is thought to entail cognitive inhibition, could be modulated by systemically administered GABAergic pharmacons in non-human primates.Twelve healthy adult rhesus macaques had been extensively trained in the PAL object-location working memory task on the Monkey CANTAB touchscreen test device. In each trial of the PAL task, the animals had to recall the locations of 3-7 distinct sequentially presented schemati

Disruption in the excitatory and inhibitory (E/I) balance of cortical networks is well-characterised in age-related neurocognitive disorders (NCD), impacting functional cognitive decline. Thus, utilising non-invasive transcranial magnetic stimulation (TMS) and electroencephalography (EEG) methods, similarly applicable in both patients and translationally relevant preclinical animal models, we established baseline cortical excitability in rhesus macaques, followed by modulation of the E/I balance by diazepam (GABAA PAM).We recorded scalp-EEG from 27 electrodes by a telemetric amplifier system d

Seizure-like activity (SLA) results from synchronous neuronal activity in the mouse hippocampal subregions, namely: CA1, CA3 and DG. While many studies have proved the anticonvulsant properties of the administration of anti-epileptic drugs, such as diazepam (DZP), a GABA-A receptor agonist, and carbamazepine (CBZ), a sodium channel blocker, their role in modulating the oscillatory patterns within these three regions remains unclear. Furthermore, the role of oscillatory dynamics in the pre-ictal period in predicting the emergence of a seizure events require further investigation. In our study,
