Effect of enriched environment and aripiprazole on selected behavioral outcomes and neurogenic markers in animal model of PTSD

Jana Osacka, Lila Dziewiczova, Lubica Horvathova, Andrej Tillinger · FENS Forum 2024

Post-traumatic stress disorder (PTSD) can develop after exposure to severe traumatic event. PTSD is associated with changes in behavior and reduced neurogenesis. Available data suggests that antipsychotics or enriched environment may positively affect behavioral outcomes and increase neurogenesis in PTSD patients. Our aim was to determine whether aripiprazole, enriched environment or their combination in the PTSD animal model will positively affect behavioral outcomes and hippocampal neurogenesis. We used the single prolonged stress paradigm to induce PTSD related symptoms in animals. Animals

Exploring the potential of induced neural stem cells (iNSCs) as therapy for spinal cord injury in a rat model

Ibrahim Khan, Lara Bieler, Katharina Günther, Dominika Jakubec-Haščák, Felix Hübl, Jaqueline Illek, Marta Suarez-Cubero, Frank Edenhofer, Sebastien Couillard-Despres · FENS Forum 2024

Spinal cord injury (SCI) is a severe condition associated with high morbidity and mortality rates. As of now, no effective therapies are available. However, induced neural stem cells (iNSCs) offer a promising therapeutic approach by enabling autologous transplantation. In the subchronic phase 30 days post-SCI, we conducted the transplantation of fGFP-labelled human iNSCs (or vehicle) into female Fischer-344 rats with moderate to severe spinal cord injuries at the T8 level. One day prior to cell transplantation, immunosuppressive treatment was started and maintained to prevent graft rejection th

Immunohistochemical characterization of the developing naked mole rat brain

Niamh Carr, Firdevs Murad, Alison J Barker · FENS Forum 2024

Naked mole rats are highly vocal eusocial mammals, that rely heavily on vocal communication to form and maintain social groups. We are interested in researching how vocal development genes and neurogenesis markers are expressed in the naked mole rat brain at different developmental timepoints. Here we report preliminary data on immunohistochemical characterization of cortical expression of FOXP2 and Doublecortin in the naked mole rat brain. FOXP2, forkhead-box protein P2, is a transcription factor associated with speech and language. Doublecortin is a neurogenesis marker that shows newly gener

Metformin administration mitigates early life stress-induced disruptions in hippocampal neurogenesis

Ekin Baysal, İrem İnanç, Deniz Billur, Esra Erdemli · FENS Forum 2024

Early Life Stress (ELS) can result in enduring health consequences, highlighting the need for exploring timely intervention approaches. Our study aims to investigate whether metformin, a long-recognized diabetes medication with neuroprotective and neurotrophic potential, can mitigate ELS-induced disruptions in hippocampal neurogenesis and dendritic growth. To test this hypothesis, both male and female neonatal Wistar rats were allocated into four distinct groups: control, metformin treatment, ELS model, and ELS model with metformin treatment (ELS+M). During the initial two weeks, ELS model was

Neuronal signature of spatial memory in the hippocampus of homing pigeons

Marie Ziegler, Guillermo Hidalgo Gadea, John Tuff, Masahiro Inda, Roland Pusch, Tobias Otto, Jonas Rose, Onur Güntürkün, Noemi Rook · FENS Forum 2024

Spatial cognition is a field of research that investigates the basis of human and animal navigation and orientation. Many animals possess excellent spatial abilities like homing and migration that can be found across the animal kingdom. The hippocampus is an essential brain area for memory formation and spatial cognition in both mammals and birds, which has been shown physiologically in the form of place and head-direction cells as well as functionally through for example ablation studies. However, in contrast to mammals, the precise pattern of hippocampal spatial processing is still rather un

Non-dividing “immature” neurons in subcortical brain regions of mammals display phylogenetic variation with clear prevalence in primates

Marco Ghibaudi, Nikita Telitsyn, Jean-Marie Graïc, Irmgard Amrein, Chet C. Sherwood, Luca Bonfanti · FENS Forum 2024

Brain structural plasticity is heterogeneous in mammals, due to adaptation to ecological pressures. Whether the genesis of new neurons (stem cell-driven adult neurogenesis) is reduced from mice to humans, we recently demonstrated that a population of prenatally generated (non-dividing) “immature” neurons (INs) retaining the expression of immaturity markers (e.g., doublecortin; DCX), is highly abundant in the cerebral cortex of gyrencephalic species. Presence of DCX-positive (DCX+) cells has been reported also in subcortical regions of some mammals, yet the results regarding their origin are co

Synergistic effects of intranasally administered GALR2 and Y1R agonists on cognitive and mood-related behaviors in adult rats: Implications for neurodegenerative and mood disorders

Manuel Narvaez Pelaez, Isabel Moreno Madrid, Jose Carlos Arrabal Gómez, Pedro Serrano castro, Estela Diaz Sanchez, Jose Erik Alvarez Contino, Miguel Angel Barbancho Fernández, Jose Andrés Sánchez Pérez, Encarnación Blanco Reina, Kjell Fuxe, Dasiel O. Borroto Escuela, Natalia García Casares · FENS Forum 2024

Background: The complexity of neurodegenerative and mood disorders necessitates innovative therapeutic strategies beyond current treatments. This study explores the combined therapeutic potential of Galanin receptor 2 (GALR2) and Neuropeptide Y Y1 receptor (Y1R) agonists, administered intranasally, on spatial memory, antidepressant-like behavior, and neurogenesis in adult rats. Methods: The investigation employed intranasal co-delivery of GALR2 agonist M1145 and Y1R agonist to adult rats, examining spatial memory through the object-in-place task and antidepressant-like effects via the Forced Sw
