Centrally located transient receptor potential ankyrin 1 ion channel may exert diverse impact on the symptoms of posttraumatic stress disorder in mice

János Konkoly, Viktória Kormos, Balázs Gaszner, Dóra Zelena, Erika Pintér · FENS Forum 2024

Introduction: Transient receptor potential ankyrin 1 (TRPA1) is a non-selective cation channel expressed in the urocortin 1 (UCN1) positive neurons of the centrally projecting Edinger-Westphal nucleus (EWcp) having modulatory impact on stress adaptation. As post-traumatic stress disorder (PTSD) is triggered by disturbed stress adaptation, we hypothesized the regulatory role of EWcp/UCN1/Trpa1 neurons in PTSD.Methods: Male TRPA1 wild-type (WT) and knockout (KO) mice were exposed to two different models of PTSD. In single prolonged stress (SPS) paradigm, time spent immobility was measured during

Involvement of peptidergic Edinger-Westphal nucleus in the neurobiology of migraine

The urocortin 1 (UCN1)-expressing neurons of the centrally projecting Edinger-Westphal nucleus (EWcp) are affected by the circadian rhythm, hormonal changes, stress and pain that are known to trigger migraine. Here we aimed at investigating the possible role of EWcp in migraine neurobiology.RNAscope in situ hybridization (ISH) combined with immunostaining was used to examine the expression of calcitonin gene-related peptide (CGRP) receptor component (Crcp) in both mouse and human EWcp and dorsal raphe nucleus (DRN). Anterograde and retrograde tracing study investigated the neuroanatomical conn

Specialized secretory neurons in the hypothalamus and the Edinger-Westphal nucleus secrete severalneurohormones. Very little is known about the regulation of their expression at the post-transcriptionallevel. Here we discovered that a non-canonical cytoplasmic poly(A) polymerase Tent5a is a globalpositive regulator of neurohormone expression. We detect Tent5a in Edinger-Westphal, paraventricularand supraoptic nuclei, as well as the lateral hypothalamic area. Importantly, RNA sequencing of theseregions’ biopsies, as well as northern analysis of the whole hypothalami, revealed decreased expressi
