Elevated synaptic pruning in microglia across patient-derived brain organoids

Susmita Malwade, Samudyata Samudyata, Marja Koskuvi, Jessica Gracias Lekander, Markus Storvik, Jari Tiihonen, Jari Koistinaho, Carl Sellgren · FENS Forum 2024

Schizophrenia (SCZ) is a highly heritable psychiatric disorder with disabling psychotic and cognitive symptoms and existing treatments are only partially effective in restoring function. Given the extensive elimination of synapses by microglia in the human cerebral cortex during early development, it has been proposed that excessive synaptic pruning may contribute to the observed reduction in gray matter thickness and synapse density in SCZ patients. We modelled synaptic pruning using patient-derived stem-cell-based forebrain organoids, containing innately developed microglia, and assessed mic

The role of TAp73 in neurodevelopment

Karina Violou, Yotam Menuchin-Lasowski, Thomas Rauen, Hans R. Schöler, Heymut Omran, Julia Wallmeier · FENS Forum 2024

The transcription factor P73 belongs to the p53 protein family, and its gene encodes two main isoforms: the transcriptionally active TAp73 isoform and the dominant-negative ΔNp73. Both isoforms play pivotal roles in regulating the self-renewal and differentiation of neural stem/progenitor cells, thus play a critical role in brain development during neuronal differentiation. Mutations in TP73 are associated with lissencephaly in humans and severe brain defects in mice. The role of TP73 and its isoforms during neurogenesis in human is still unclear. Aiming to further scrutinize the role of TAP73
