Sensory stimulation boosts brain dynamics fluidity and memory performance in Alzheimer’s disease mice

Demian Battaglia, Matthieu Aguilera, Samy Castro, Jyotika Bahuguna, Laura Harsan, Chantal Mathis, Romain Goutagny · COSYNE 2025

Visual gamma entrainment using sensory stimuli (vGENUS) is a promising non-invasive therapeutic approach for Alzheimer's disease (AD), showing efficacy in improving memory function and physiological biomarkers levels in mice and, in perspective, also human patients. However, its mechanisms of action remain poorly understood. Explanations in terms of boosted neurogenesis or glymphatic clearance have been advanced. Here we probe the alternative hypothesis that this non-invasive and non-pharmacological stimulation acts by increasing the fluidity of brain dynamics. Our hypothesis expands upon prev

40 Hz gamma visual stimulation restores brain dynamics and boosts memory in a new mouse model of early Alzheimer’s disease

Matthieu Aguilera, Chantal Mathis, Demian Battaglia, Romain Goutagny · FENS Forum 2024

Alzheimer’s disease (AD) is a neurodegenerative disease and the world leading cause of dementia, yet still no effective treatments are available. Recently, a non-invasive therapy based on 40 Hz gamma stimulation using sensory stimuli (GENUS) showed beneficial effects on memory performances in several AD mouse models and mild probable AD patients. However, mechanisms underlying these positive effects, such as amyloid plaque clearance, microglial activation or gamma rhythm restauration, are still matter of intense debate. Here, we test the alternative hypothesis that GENUS might rescue memory de

High-density EEG in the pilocarpine model of temporal lobe epilepsy: A power spectrum analysis of tonic-clonic seizures

Beatrice Casadei Garofani, Arianna Capodiferro, Stefania Bartoletti, Federica Raimondi, Elisa Ren, Daniela Gandolfi, Giulia Curia · FENS Forum 2024

Temporal lobe epilepsy (TLE) is characterized by spontaneous recurrent seizures poorly controlled by antiseizure medications in over 30% of patients. The current study aims to investigate the most epileptogenic areas in a TLE animal model using high-density EEG.Sprague Dawley male rats were injected intraperitoneally with pilocarpine to induce the status epilepticus (SE) terminated 30 minutes later by ketamine/diazepam. Two weeks later animals were implanted with epidural electrodes in the frontal cortex and depth electrodes in several temporal areas. Video-EEG recordings (24h, >3 days/week

Memory generalization and overgeneralization in sleep

Ella Bar, Yuval Nir, Rony Paz · FENS Forum 2024

Overgeneralization and sleep quality disruption are major components of anxiety and post-traumatic stress disorder (PTSD). This study attempts to elucidate potential links between these elements. We designed a paradigm wherein participants learned associations between three faces and positive, negative, or neutral outcomes. Subsequently, we evaluated their generalization of these faces by examining responses to morphed faces resembling the initially associated faces before and after consolidation during daytime wakefulness or full-night sleep. Our findings reveal that post-sleep, participants
