Chromatin accessibility in oligodendrocyte precursors profiled by ATAC-seq: Neuroprotective effects of MgSO4 and 4-PBA alone or associated in a mouse model of encephalopathy of prematurity

Cerebral palsy (CP) is the leading cause of motor disability in children. Hypoxia-ischemia (HI) and prematurity are main risk factors. Administration of magnesium sulfate (MgSO4) to mothers before 33 weeks' amenorrhea (SA) reduces the occurrence of CP at 2 years by around 30%. At this age, the white matter composed by oligodendrocytes, is particularly vulnerable. However, at long term, MgSO4 neuroprotective effects are not significant.Given the anti-inflammatory and antioxidant properties of 4-phenylbutyrate (4-PBA), its association with MgSO4 may optimize and prolong this neuroprotection in p

Our previous study found that ectomesenchymal stem cells derived from human pluripotent stem cells (hPSC-EMSCs) exhibited higher neuroprotective potential than hUC-MSCs, as evidenced by a more significant reduction in lesion size and apoptosis in rat brains after hypoxia-ischemia (HI). Moreover, in vitro studies demonstrated that hPSC-EMSC-derived conditioned medium (CM) had neuroprotective and neurorestorative effects through the promotion of anti-apoptosis, neurite growth and neurogenesis. In this study, we discovered that hPSC-EMSCs and hUC-MSCs expressed differential secreted factors by ut

Oxytocin decreases neuroinflammation after neonatal hypoxia-ischemia

Gabriel Schirmbeck, Eduardo Farias Sanches, Stéphane Sizonenko, Olivier Baud · FENS Forum 2024

Hypoxia-ischemia (HI) is a significant cause of disability and mortality in neonates. HI is characterized by acute energetic failure and secondary inflammatory response. The primary therapeutic intervention is hypothermia, which presents limited neuroprotective effects. Environmental approaches, such as skin-to-skin contact, have been highlighted for their benefits in brain development. Oxytocin (OT) is a neuropeptide with roles in parturition and social behavior, but it also modulates the microglial inflammatory responses. Therefore, the study of OT signaling can be helpful to bridge the gap
