Cortical circuits for context dependent sensory processing

Pol Bech Vilaseca, Robin Dard, Sylvain Crochet, Carl CH Petersen · FENS Forum 2024

In nature, humans and animals are constantly bombarded with sensory information, which needs to be promptly contextualized to elicit the appropriate motor sequences through a process known as sensorimotor transformation. While we begin to understand how sensory information is transformed into motor action, it is still unclear in what way contextual information can modify a given sensorimotor plan. In this project, we investigate this phenomenon by training water restricted mice in a context-dependent multisensory detection task. We developed a behavioural paradigm in which mice are required to

Dendritic imaging of voltage and calcium signals during visual learning paradigm

Anna Jasz, Borbála Kertész, Helén Bartók, Tamás Tompa, Zoltán Szadai, Balázs Rózsa · FENS Forum 2024

Although dendritic signaling strongly influences sensory processing and learning, the connection between dendritic integration and somatic output is still not fully understood. Our aim is maping the origins of dendritic events during different aspects of visual learning. We used 3D two-photon laser scanning microscopy with acousto-optic deflectors to simultaneously record jRGECO1a (calcium) and JEDI-2p (voltage) signals in the apical dendrites of layer 5 pyramidal cells in the primary visual cortex of mice during visual discrimination. We examined mice with controlled water access participating

Mapping synaptic integration with simultaneous glutamate and calcium imaging

Federico Rossi, Anyi Liu, Kenneth D. Harris, Matteo Carandini · FENS Forum 2024

A key question in neuroscience is how neurons compute their specialized output from their synaptic inputs. Another key question is whether this computation is shaped by non-linear dendritic integration. To answer these questions, it would be ideal to map synaptic activity while recording dendritic and somatic responses. We developed a strategy to simultaneously measure glutamate synaptic release and postsynaptic responses in L2/3 pyramidal neurons in the mouse primary visual cortex. We sparsely coexpressed two sensors: the green membrane sensor iGluSnFR3, reporting presynaptic release of gluta
