Neuromodulation with DBS and FUS: Ephys effects in an animal model of depression

Lisa Ratz, Sofia Drakopoulou, Artur Fornol, Georgios Spyropoulos, Volker Arnd Coenen, Máté Daniel Döbrössy · FENS Forum 2024

Depression is one of the most common psychiatric disorders with about one third of patients being considered treatment-resistant. Various neuromodulation approaches have been explored, including deep brain stimulation (DBS) of the medial forebrain bundle (mfb), which has shown promising results in clinical studies. In recent years transcranial focused ultrasound (FUS) at low intensities has been emerging as another form of stimulation. However, challenges arise from skull absorption, necessitating lower frequencies and therefore resolutions. To address this, we report on the first steps in a co

Protective effects of intracranial stimulation on spatial memory and changes in miRNA serum levels in a sporadic rat model of Alzheimer disease: A longitudinal study

Andrea Riberas Sánchez, Soleil Garcia Brito, Gemma Carreras Badosa, Laia Vila Solés, Laura Aldavert Vera, Pilar Segura Torres, Gemma Huguet Blanco, Elisabet Kádár Garcia · FENS Forum 2024

Alzheimer’s disease (AD) is the most prevalent ageing-associated neurodegenerative disease. Deep brain stimulation is a novel therapeutic approach, but long-term effects remain unknown. We previously demonstrated that acute intracranial self-stimulation (ICSS) in medial forebrain bundle (MFB) improves the acquisition but not 72-hour retention of a spatial learning task and regulates expression of specific miRNAs in a rat AD model. This study aims to analyse the effect of a chronic MFB-ICSS treatment on spatial memory and miRNA serum expression levels in a rat AD model by streptozotocin (STZ) i

Deep brain stimulation (DBS) targeting the medial forebrain bundle (MFB) is emerging as a promising intervention for treatment-resistant depression. Our study systematically investigated the NA system in a depression model and explored the modulatory effects of mfb-DBS on this system.Using Flinders Sensitive Line (FSL) rats as a depression model and Sprague Dawley (SD) rats as controls, we examined the spatial distribution and myelination of NA fibers within the mfb under confocal microscopy, monitored real-time in vivo NA release in response to mfb-DBS in the mPFC and NAC during 6 different s

Sleep-spindle triggered closed-loop neuromodulation of pathological anxiety in rats

Andrea Pejin, Lívia Barcsai, Nóra Forgó, Rodrigo Sierra, Lizeth Pedraza, Olivér Nagy, Magor L. Lőrincz, Antal Berényi · FENS Forum 2024

Post-traumatic stress disorder affects millions worldwide, with low therapeutic success. Research confirms the involvement of memory processes in pathological and persistent traumatic memories that are difficult to overwrite. Previously, in an animal model, we demonstrated that the extinction can be expedited by closed-loop (CL) stimulation of the medial forebrain bundle (MFB) triggered by sharp-wave ripple (SWR) patterns. Similarly, stimulation of the infralimbic cortex (IL) timed to SWR also maintained a low level of fear response over the long term.We aimed to investigate if the manipulatio
