Lactate influences the m6A RNA methylation of genes involved in neuronal function

Fathia Ben-Rached, Anel Lopez-Gonzalez, Gabriel Herrera-Lopez, John S. Girgis, Pierre J. Magistretti, Hubert Fiumelli · FENS Forum 2024

The Astrocyte-Neuron Lactate Shuttle model posits that lactate is transferred from astrocytes to neurons to fulfill energetic needs and act as a signal that regulates gene expression related to neuroprotection, neuronal excitability, synaptic plasticity, and memory. N6-methyladenosine (m6A), the most abundant internal modification of RNA, influences mRNA stability, translation, transport, localization, and alternative splicing. This dynamic epitranscriptomic mark is prevalent in the nervous system, impacting various processes, including those affected by lactate, such as neurogenesis, synaptic

Repetitive transcranial magnetic stimulation induced changes of N6-methyladenosine modified mRNAs

Elli-Anna Balta, Christos Galanis, Jakob Straehle, Michael Uhl, Matthias Kirsch, Rolf Backofen, Juergen Beck, Andreas Vlachos · FENS Forum 2024

Repetitive transcranial magnetic stimulation (rTMS) is a widely used method of non-invasive brain stimulation in both research and clinical settings, aimed at inducing synaptic plasticity and targeting conditions like pharmaco-resistant depression and post-traumatic stress disorder. However, the detailed molecular mechanisms underlying rTMS-induced plasticity remain not well understood. N6-methyladenosine (m6A) is the most prevalent modification in the human transcriptome. m6A is known for its reversible nature and thus is a prime candidate for modulating synaptic plasticity via gene expressio

Uncovering transcriptome-wide lncRNA methylation and expression patterns in human glioma

Rugile Dragunaite, Giedrius Steponaitis, Rytis Stakaitis, Daina Skiriute · FENS Forum 2024

Alterations of N6-methyladenosine (m6A) RNA modifications are responsible for a platter of structural and functional changes in healthy and cancerous cell states. Even though several studies have linked m6A to the onset and development of tumors, very few have examined transcriptome-wide m6A landscape of long non-coding RNAs (lncRNAs) in cancerous cells, including gliomas. This work aims to explore and elucidate lncRNA m6A alterations and expression in glioma stem cells, U87-MG cells and glioma patients. We utilized four cell lines (NCH421k, NCH644, NCH690, U-87 MG), along with 16 glioblastoma
