Influence of serotonergic modulation in recovery after stroke

Amido Daugardt, Leif Koschützke, Livia Wieland, Nikolaus Wenger, Christoph Harms · FENS Forum 2024

Current rehabilitation for stroke primarily relies on physiotherapy. Research has found that serotonin plays a crucial role in the neuroplastic processes required for brain reorganisation after a stroke. We investigated whether modulation of serotonergic transmission of the dorsal raphe nucleus (DRN), together with rehabilitation training, leads to an improvement in motor recovery. In a photothrombotic stroke mouse model, lesion volume and localization were assessed following 24h after ischemia with 7T-MRI. We chemogenetically activated or inhibited the DRN through adeno-associated virus (AAV)

Ischemic stroke results from neural tissue death due to a decrease in blood supply, and is a major cause of death and motor disabilities worldwide. Although it is established that the tissue immediately surrounding the stroke core undergoes spontaneous functional reorganization, partially taking over lost functionality, the role of the healthy contralesional cortex remains unclear. Adaptions in neuronal activity on the contralesional side have been reported both in humans and in rodent stroke models, but it remains elusive whether such modifications represent spontaneous functional re-mapping
