Extracellular vesicles: An exploration into the bi-directional crosstalk of endothelial cells and astrocytes at the blood-brain barrier

Natalija Aleksejenko, Keith D. Rochfort, Philip M. Cummins, Janosch P. Heller · FENS Forum 2024

Endothelial cells (ECs) tightly regulate the passage of ions, molecules, and cells from the blood into the brain and vice versa as part of the blood-brain barrier (BBB). Astrocytes aid in the formation and maintenance of the BBB. Aside from direct communication, astrocytes and ECs also release extracellular vesicles (EVs) to communicate with each other and other cells. EVs carry genetic information, proteins, and other signalling molecules. This cargo changes relative to the donor cell state, which can influence different processes in recipient cells. This project investigates this bi-direction

Impact of perinatal exposure to maternal western diet on offspring socioemotional behavior

Jerome Becker, Pauline Monguillon, Agathe Brugoux, Mélanie Morin, Enola Roussin, Sylviane Marouillat, Patrick Vourch’, Eduardo Gascon, Laetitia Davidovic, Julie Le Merrer · FENS Forum 2024

Maternal diet is a key determinant of later child’s metabolic and mental health outcomes. Human and/or animals consuming a western diet (WD), as compared to a healthy diet (HD), exhibit microbiota dysbiosis, low-grade inflammation, behavioral alterations and defects in the central reward circuit. Accordingly, epidemiological studies show that WD consumption during pregnancy is associated with adverse metabolic and mental health outcomes in offspring. We have developed a mouse model to study the impact of maternal WD on the offspring and challenged hypotheses related to dysbiosis, inflammation

Mechanistic single-cell investigation of neuroinflammation induced by influenza A virus infection

Lea Gabele, Shirin Hosseini, Kristin Michaelsen-Preusse, Nele Rieke, Christian Sieben, Martin Korte · FENS Forum 2024

Influenza A virus (IAV) infection can lead to cerebral manifestations, potentially increasing the risk for neurodegenerative diseases. Some IAV strains, such as avian H7N7 subtype, are neurotropic and capable of invading and infecting resident cells in the brain. IAV-induced production of proinflammatory cytokines leads to activation of microglia, thereby potentially triggering neuronal damage and dysfunction. Microglial characteristics such as phagocytosis rate, activity and morphology are sex-dependent. Therefore, a better understanding of IAV neurotropism and the mechanisms leading to delet

P. gingivalis and titanium oxide nanoparticles induce inflammation in BBB microglia in vitro

Yasmin Louzon, Rinat Tzach-Nachman, Yael Houri-Haddad · FENS Forum 2024

Research has extensively explored the influence of Periodontitis on Alzheimer’s disease (AD), identifying its role in accelerating the disease via systemic inflammation and bacterial invasion to the brain. Additionally, investigations into titanium oxide nanoparticles (Ti NP) and AD have shown significant effects on cognitive ability in vitro and in vivo. This study aims to assess the synergistic impact of Ti NP and P. gingivalis (Pg), factors involved in periimplantitis pathogenesis, on the neuroinflammatory response of mouse microglia cells (BV2) in vitro.Using a BV2 and RAW (separate contro
