High precision ultrasound stimulation of the retina with photoacoustic membrane

Audrey Leong, Yueming Li, Julien Voillot, Arnaud Facon, Chakrya-Anna Chhuon, Clémence Bradic, Jean-Damien Louise, Serge Rosolen, Hélène Moulet, Chen Yang, Ji-Xin Cheng, Serge Picaud · FENS Forum 2024

Ultrasound stimulation of the retina was proposed as an alternative to retinal prostheses for visual restoration. Prior works have shown that the retina responds to mechanical stimulation, but no attempt on stimulating an ex vivo retina with a free-standing photoacoustic membrane had yet been made. Here, we report ultrahigh precision stimulation of the retina with optically generated focused ultrasound (OFUS). OFUS is generated with a photoacoustic membrane of embedded carbon nanoparticles in polydimetylsiloxane. Using ex vivo multielectrode array recording and pharmalogical approaches, we inv

Retinotopic characterization of primate visual cortex using functional ultrasound imaging for sonogenetic stimulation

Ignacio Alcala, Matthieu Provansal, Fabrice Arcizet, Serge Picaud · FENS Forum 2024

In a recent study in rodents in our team, we showed that sonogenetic therapy can be applied to stimulate the visual cortex (V1) at a high spatiotemporal resolution with ultrasound following the expression of the MscL mechanosensitive ionic channel. Moreover, we proved that sonogenetic activation of V1 can generate visual perception using an associative learning test.To develop sonogenetics as a vision restoration therapy the next step is to prove visual perception via sonogenetic stimulation in non-human primates (NHPs).Methods: In consequence, we proceeded to obtain retinotopic maps of NHP’s

Sonogenetics: From the mechano-sensitive channel to brain stimulation for visual restoration

Erwan Dessailly, Ignacio Alcala, Matthieu Provencal, Audrey Leong, Rafik Arab, Pierre Pouget, Fabrice Arcizet, Serge Picaud · FENS Forum 2024

In the pursuit of restoring vision for patients having lost the eye-brain connection, ultrasound stimulation is a new interesting therapeutic approach at the cortical level. In particular, Sonogenetic therapy has been explored as a mean to increase the sensitivity of cells to ultrasound by expressing a mechanosensitive ion channel in the membranes of cells of interest in order to modulate their activity. In order to optimize sonogenetics for vision restoration, we first focused on exploring the properties of different mechano-sensitive ion channels, the key parameters being the spatial and temp
