Dual inhibition of ecto-5'-nucleotidase (CD73) and adenosine A2A receptor reduces neuroinflammation and oxidative stress in TNF, IL-1α, C1q-induced neurotoxic astrocytes

Ecto-5'-nucleotidase (CD73) is an ecto-enzyme that hydrolyzes adenosine monophosphate to adenosine, which acts through one of four subtypes of adenosine receptors. Increased CD73 activity and expression in chronic neurodegenerative diseases leads to a higher concentration of adenosine in the extracellular space. This results in excessive activation of adenosine A2AR, which triggers proinflammatory signaling pathways that further promote neurodegeneration. Therefore, dual CD73/A2AR inhibition is a potential pharmacologic target in chronic neurodegeneration. In the present study, we used a novel

Unravelling the mechanisms behind development of quiescent neural stem cells: The role of Presenilin 1

Surya Suresh, Jyothi Pradeep Nair, Meera Vadakkath, Riya Paul Ann, Parvathy Surendran, Rahul Jose, Swathy Vankudoth, Avyanga Sankaranarayanan, Jackson James · FENS Forum 2024

During embryonic development, some of the proliferating neural stem cells enter a quiescent state and is retained till adulthood, thus contributing to adult neurogenesis, a process involving different proteins. From a published microarray of Presenilin-1 knock out embryo, one such protein, VCAM1 was found to be significantly downregulated. In addition, Presenilin-1(PS1) is known to be involved in embryonic neurogenesis as well as in cell cycle regulation. Based on these, we hypothesized PS1 to have a role in neural stem cell quiescence. To address this, we modelled quiescence in vitro by additi
