“Brain theory, what is it or what should it be?”
n the neurosciences the need for some 'overarching' theory is sometimes expressed, but it is not always obvious what is meant by this. One can perhaps agree that in modern science observation and experimentation is normally complemented by 'theory', i.e. the development of theoretical concepts that help guiding and evaluating experiments and measurements. A deeper discussion of 'brain theory' will require the clarification of some further distictions, in particular: theory vs. model and brain research (and its theory) vs. neuroscience. Other questions are: Does a theory require mathematics? Or even differential equations? Today it is often taken for granted that the whole universe including everything in it, for example humans, animals, and plants, can be adequately treated by physics and therefore theoretical physics is the overarching theory. Even if this is the case, it has turned out that in some particular parts of physics (the historical example is thermodynamics) it may be useful to simplify the theory by introducing additional theoretical concepts that can in principle be 'reduced' to more complex descriptions on the 'microscopic' level of basic physical particals and forces. In this sense, brain theory may be regarded as part of theoretical neuroscience, which is inside biophysics and therefore inside physics, or theoretical physics. Still, in neuroscience and brain research, additional concepts are typically used to describe results and help guiding experimentation that are 'outside' physics, beginning with neurons and synapses, names of brain parts and areas, up to concepts like 'learning', 'motivation', 'attention'. Certainly, we do not yet have one theory that includes all these concepts. So 'brain theory' is still in a 'pre-newtonian' state. However, it may still be useful to understand in general the relations between a larger theory and its 'parts', or between microscopic and macroscopic theories, or between theories at different 'levels' of description. This is what I plan to do.
Mapping the neural dynamics of dominance and defeat
Social experiences can have lasting changes on behavior and affective state. In particular, repeated wins and losses during fighting can facilitate and suppress future aggressive behavior, leading to persistent high aggression or low aggression states. We use a combination of techniques for multi-region neural recording, perturbation, behavioral analysis, and modeling to understand how nodes in the brain’s subcortical “social decision-making network” encode and transform aggressive motivation into action, and how these circuits change following social experience.
Unmotivated bias
In this talk, I will explore how social affective biases arise even in the absence of motivational factors as an emergent outcome of the basic structure of social learning. In several studies, we found that initial negative interactions with some members of a group can cause subsequent avoidance of the entire group, and that this avoidance perpetuates stereotypes. Additional cognitive modeling discovered that approach and avoidance behavior based on biased beliefs not only influences the evaluative (positive or negative) impressions of group members, but also shapes the depth of the cognitive representations available to learn about individuals. In other words, people have richer cognitive representations of members of groups that are not avoided, akin to individualized vs group level categories. I will end presenting a series of multi-agent reinforcement learning simulations that demonstrate the emergence of these social-structural feedback loops in the development and maintenance of affective biases.

Decomposing motivation into value and salience
Humans and other animals approach reward and avoid punishment and pay attention to cues predicting these events. Such motivated behavior thus appears to be guided by value, which directs behavior towards or away from positively or negatively valenced outcomes. Moreover, it is facilitated by (top-down) salience, which enhances attention to behaviorally relevant learned cues predicting the occurrence of valenced outcomes. Using human neuroimaging, we recently separated value (ventral striatum, posterior ventromedial prefrontal cortex) from salience (anterior ventromedial cortex, occipital cortex) in the domain of liquid reward and punishment. Moreover, we investigated potential drivers of learned salience: the probability and uncertainty with which valenced and non-valenced outcomes occur. We find that the brain dissociates valenced from non-valenced probability and uncertainty, which indicates that reinforcement matters for the brain, in addition to information provided by probability and uncertainty alone, regardless of valence. Finally, we assessed learning signals (unsigned prediction errors) that may underpin the acquisition of salience. Particularly the insula appears to be central for this function, encoding a subjective salience prediction error, similarly at the time of positively and negatively valenced outcomes. However, it appears to employ domain-specific time constants, leading to stronger salience signals in the aversive than the appetitive domain at the time of cues. These findings explain why previous research associated the insula with both valence-independent salience processing and with preferential encoding of the aversive domain. More generally, the distinction of value and salience appears to provide a useful framework for capturing the neural basis of motivated behavior.
Prosocial Learning and Motivation across the Lifespan
2024 BACN Early-Career Prize Lecture Many of our decisions affect other people. Our choices can decelerate climate change, stop the spread of infectious diseases, and directly help or harm others. Prosocial behaviours – decisions that help others – could contribute to reducing the impact of these challenges, yet their computational and neural mechanisms remain poorly understood. I will present recent work that examines prosocial motivation, how willing we are to incur costs to help others, prosocial learning, how we learn from the outcomes of our choices when they affect other people, and prosocial preferences, our self-reports of helping others. Throughout the talk, I will outline the possible computational and neural bases of these behaviours, and how they may differ from young adulthood to old age.
Neural mechanisms governing the learning and execution of avoidance behavior
The nervous system orchestrates adaptive behaviors by intricately coordinating responses to internal cues and environmental stimuli. This involves integrating sensory input, managing competing motivational states, and drawing on past experiences to anticipate future outcomes. While traditional models attribute this complexity to interactions between the mesocorticolimbic system and hypothalamic centers, the specific nodes of integration have remained elusive. Recent research, including our own, sheds light on the midline thalamus's overlooked role in this process. We propose that the midline thalamus integrates internal states with memory and emotional signals to guide adaptive behaviors. Our investigations into midline thalamic neuronal circuits have provided crucial insights into the neural mechanisms behind flexibility and adaptability. Understanding these processes is essential for deciphering human behavior and conditions marked by impaired motivation and emotional processing. Our research aims to contribute to this understanding, paving the way for targeted interventions and therapies to address such impairments.
Computational and mathematical approaches to myopigenesis
Myopia is predicted to affect 50% of all people worldwide by 2050, and is a risk factor for significant, potentially blinding ocular pathologies, such as retinal detachment and glaucoma. Thus, there is significant motivation to better understand the process of myopigenesis and to develop effective anti-myopigenic treatments. In nearly all cases of human myopia, scleral remodeling is an obligate step in the axial elongation that characterizes the condition. Here I will describe the development of a biomechanical assay based on transient unconfined compression of scleral samples. By treating the scleral as a poroelastic material, one can determine scleral biomechanical properties from extremely small samples, such as obtained from the mouse eye. These properties provide proxy measures of scleral remodeling, and have allowed us to identify all-trans retinoic acid (atRA) as a myopigenic stimulus in mice. I will also describe nascent collaborative work on modeling the transport of atRA in the eye.
Obesity and Brain – Bidirectional Influences
The regulation of body weight relies on homeostatic mechanisms that use a combination of internal signals and external cues to initiate and terminate food intake. Homeostasis depends on intricate communication between the body and the hypothalamus involving numerous neural and hormonal signals. However, there is growing evidence that higher-level cognitive function may also influence energy balance. For instance, research has shown that BMI is consistently linked to various brain, cognitive, and personality measures, implicating executive, reward, and attentional systems. Moreover, the rise in obesity rates over the past half-century is attributed to the affordability and widespread availability of highly processed foods, a phenomenon that contradicts the idea that food intake is solely regulated by homeostasis. I will suggest that prefrontal systems involved in value computation and motivation act to limit food overconsumption when food is scarce or expensive, but promote over-eating when food is abundant, an optimum strategy from an economic standpoint. I will review the genetic and neuroscience literature on the CNS control of body weight. I will present recent studies supporting a role of prefrontal systems in weight control. I will also present contradictory evidence showing that frontal executive and cognitive findings in obesity may be a consequence not a cause of increased hunger. Finally I will review the effects of obesity on brain anatomy and function. Chronic adiposity leads to cerebrovascular dysfunction, cortical thinning, and cognitive impairment. As the most common preventable risk factor for dementia, obesity poses a significant threat to brain health. I will conclude by reviewing evidence for treatment of obesity in adults to prevent brain disease.
Cognition in the Wild
What do nonhuman primates know about each other and their social environment, how do they allocate their attention, and what are the functional consequences of social decisions in natural settings? Addressing these questions is crucial to hone in on the co-evolution of cognition, social behaviour and communication, and ultimately the evolution of intelligence in the primate order. I will present results from field experimental and observational studies on free-ranging baboons, which tap into the cognitive abilities of these animals. Baboons are particularly valuable in this context as different species reveal substantial variation in social organization and degree of despotism. Field experiments revealed considerable variation in the allocation of social attention: while the competitive chacma baboons were highly sensitive to deviations from the social order, the highly tolerant Guinea baboons revealed a confirmation bias. This bias may be a result of the high gregariousness of the species, which puts a premium on ignoring social noise. Variation in despotism clearly impacted the use of signals to regulate social interactions. For instance, male-male interactions in chacma baboons mostly comprised dominance displays, while Guinea baboon males evolved elaborate greeting rituals that serve to confirm group membership and test social bonds. Strikingly, the structure of signal repertoires does not differ substantially between different baboon species. In conclusion, the motivational disposition to engage in affiliation or aggressiveness appears to be more malleable during evolution than structural elements of the behavioral repertoire; this insight is crucial for understanding the dynamics of social evolution.

Analyzing artificial neural networks to understand the brain
In the first part of this talk I will present work showing that recurrent neural networks can replicate broad behavioral patterns associated with dynamic visual object recognition in humans. An analysis of these networks shows that different types of recurrence use different strategies to solve the object recognition problem. The similarities between artificial neural networks and the brain presents another opportunity, beyond using them just as models of biological processing. In the second part of this talk, I will discuss—and solicit feedback on—a proposed research plan for testing a wide range of analysis tools frequently applied to neural data on artificial neural networks. I will present the motivation for this approach as well as the form the results could take and how this would benefit neuroscience.
Analogy Use in Parental Explanation
How and why are analogies spontaneously generated? Despite the prominence of analogy in learning and reasoning, there is little research on whether and how analogy is spontaneously generated in everyday settings. Here we fill this gap by gathering parents' answers to children's real questions, and examining analogy use in parental explanations. Study 1 found that parents used analogy spontaneously in their explanations, despite no prompt nor mention of analogy in the instruction. Study 2 found that these analogical explanations were rated highly by parents, schoolteachers, and university students alike. In Study 3, six-year-olds also rated good analogical explanations highly, but unlike their parents, did not rate them higher than causal, non-analogical explanations. We discuss what makes an analogy a good explanation, and how theories from both explanation and analogy research explain one’s motivation for spontaneously generating analogies.
Chemistry of the adaptive mind: lessons from dopamine
The human brain faces a variety of computational dilemmas, including the flexibility/stability, the speed/accuracy and the labor/leisure tradeoff. I will argue that striatal dopamine is particularly well suited to dynamically regulate these computational tradeoffs depending on constantly changing task demands. This working hypothesis is grounded in evidence from recent studies on learning, motivation and cognitive control in human volunteers, using chemical PET, psychopharmacology, and/or fMRI. These studies also begin to elucidate the mechanisms underlying the huge variability in catecholaminergic drug effects across different individuals and across different task contexts. For example, I will demonstrate how effects of the most commonly used psychostimulant methylphenidate on learning, Pavlovian and effortful instrumental control depend on fluctuations in current environmental volatility, on individual differences in working memory capacity and on opportunity cost respectively.
Can I be bothered? Neural and computational mechanisms underlying the dynamics of effort processing (BACN Early-career Prize Lecture 2021)
From a workout at the gym to helping a colleague with their work, everyday we make decisions about whether we are willing to exert effort to obtain some sort of benefit. Increases in how effortful actions and cognitive processes are perceived to be has been linked to clinically severe impairments to motivation, such as apathy and fatigue, across many neurological and psychiatric conditions. However, the vast majority of neuroscience research has focused on understanding the benefits for acting, the rewards, and not on the effort required. As a result, the computational and neural mechanisms underlying how effort is processed are poorly understood. How do we compute how effortful we perceive a task to be? How does this feed into our motivation and decisions of whether to act? How are such computations implemented in the brain? and how do they change in different environments? I will present a series of studies examining these questions using novel behavioural tasks, computational modelling, fMRI, pharmacological manipulations, and testing in a range of different populations. These studies highlight how the brain represents the costs of exerting effort, and the dynamic processes underlying how our sensitivity to effort changes as a function of our goals, traits, and socio-cognitive processes. This work provides new computational frameworks for understanding and examining impaired motivation across psychiatric and neurological conditions, as well as why all of us, sometimes, can’t be bothered.
Neural Representations of Social Homeostasis
How does our brain rapidly determine if something is good or bad? How do we know our place within a social group? How do we know how to behave appropriately in dynamic environments with ever-changing conditions? The Tye Lab is interested in understanding how neural circuits important for driving positive and negative motivational valence (seeking pleasure or avoiding punishment) are anatomically, genetically and functionally arranged. We study the neural mechanisms that underlie a wide range of behaviors ranging from learned to innate, including social, feeding, reward-seeking and anxiety-related behaviors. We have also become interested in “social homeostasis” -- how our brains establish a preferred set-point for social contact, and how this maintains stability within a social group. How are these circuits interconnected with one another, and how are competing mechanisms orchestrated on a neural population level? We employ optogenetic, electrophysiological, electrochemical, pharmacological and imaging approaches to probe these circuits during behavior.
The neural basis of pain experience and its modulation by opioids
How the brain creates a painful experience remains a mystery. Solving this mystery is crucial to understanding the fundamental biological processes that underlie the perception of body integrity, and to creating better, non-addictive pain treatments. My laboratory’s goal is to resolve the neural basis of pain. We aim to understand the mechanisms by which our nervous system produces and assembles the sensory-discriminative, affective-motivational, and cognitive-evaluative dimensions of pain to create this unique and critically important experience. To capture every component of the pain experience, we examine the entirety of the pain circuitry, from sensory and spinal ascending pathways to cortical/subcortical circuits and brainstem descending pain modulation systems, at the molecular, cellular, circuit and whole-animal levels. For these studies, we have invented novel behavioral paradigms to interrogate the affective and cognitive dimensions of pain in mice while simultaneously imaging and manipulating nociceptive circuits. My laboratory also investigates how opioids suppress pain. Remarkably, despite their medical and societal significance, how opium poppy alkaloids such as morphine produce profound analgesia remains largely unexplained. By identifying where and how opioids act in neural circuits, we not only establish the mechanisms of action of one of the oldest drugs known to humans, but also reveal the critical elements of the pain circuitry for developing of novel analgesics and bringing an end to the opioid epidemic.
Free will over time: Distinguishing top-down and now-then control
Self-control is a central aspect of free will. Because self-control is often described in terms of resisting temptations, research on the cognitive neuroscience of free will often focuses on mechanisms of top-down regulation. We argue that this obscures a crucial temporal dimension of free will: now-then regulation. We distinguish now-then regulation from top-down regulation, and situate now-then regulation within a broader account of temporally extended agency. In highlighting this temporal dimension of control, we aim to provide a more nuanced account of how motivation informs action over time, different kinds of regulatory processes underlying the planning and execution of action, and the temporal components of reasons-responsiveness.
Analogy and ethics: opportunities at the intersection
Analogy offers a new interpretation of a common concern in ethics: whether decision making includes or excludes a consideration of moral issues. This is often discussed as the moral awareness of decision makers and considered a motivational concern. The possible new interpretation is that moral awareness is in part a matter of expertise. Some failures of moral awareness can then be understood as stemming from novicehood. Studies of analogical transfer are consistent with the possibility that moral awareness is in part a matter of expertise, that as a result motivation is less helpful than some prior theorizing would predict, and that many adults are not as expert in the domain of ethics as one might hope. The possibility of expert knowledge of ethical principles leads to new questions and opportunities.
Learning to aggress – Behavioral and circuit mechanisms of aggression reward
Aggression is an ethologically complex behavior with equally complex underlying mechanisms. Here, I present data on one form of aggression, appetitive or rewarding aggression, and the behavioral, cellular and system-level mechanisms guiding this behavior. First, I will present one way in which appetitive aggression is modeled in mice, and extend aggression motivation to the concept of compulsive aggression seeking and relapse. I will then briefly highlight recent advances in computer vision and machine learning for automated scoring of aggressive behavior, the role of specific cell-types in controlling aggression reward, and close with preliminary data on the whole brain aggression reward functional connectome using light sheet fluorescent microscopy (LSFM).
Differential working memory functioning
The integrated conflict monitoring theory of Botvinick introduced cognitive demand into conflict monitoring research. We investigated effects of individual differences of cognitive demand and another determinant of conflict monitoring entitled reinforcement sensitivity on conflict monitoring. We showed evidence of differential variability of conflict monitoring intensity using the electroencephalogram (EEG), functional magnet resonance imaging (fMRI) and behavioral data. Our data suggest that individual differences of anxiety and reasoning ability are differentially related to the recruitment of proactive and reactive cognitive control (cf. Braver). Based on previous findings, the team of the Leue-Lab investigated new psychometric data on conflict monitoring and proactive-reactive cognitive control. Moreover, data of the Leue-Lab suggest the relevance of individual differences of conflict monitoring for the context of deception. In this respect, we plan new studies highlighting individual differences of the functioning of the Anterior Cingulate Cortex (ACC). Disentangling the role of individual differences in working memory-related cognitive demand, mental effort, and reinforcement-related processes opens new insights for cognitive-motivational approaches of information processing (Passcode to rewatch: 0R8v&m59).
A brain circuit for curiosity
Motivational drives are internal states that can be different even in similar interactions with external stimuli. Curiosity as the motivational drive for novelty-seeking and investigating the surrounding environment is for survival as essential and intrinsic as hunger. Curiosity, hunger, and appetitive aggression drive three different goal-directed behaviors—novelty seeking, food eating, and hunting— but these behaviors are composed of similar actions in animals. This similarity of actions has made it challenging to study novelty seeking and distinguish it from eating and hunting in nonarticulating animals. The brain mechanisms underlying this basic survival drive, curiosity, and novelty-seeking behavior have remained unclear. In spite of having well-developed techniques to study mouse brain circuits, there are many controversial and different results in the field of motivational behavior. This has left the functions of motivational brain regions such as the zona incerta (ZI) still uncertain. Not having a transparent, nonreinforced, and easily replicable paradigm is one of the main causes of this uncertainty. Therefore, we chose a simple solution to conduct our research: giving the mouse freedom to choose what it wants—double freeaccess choice. By examining mice in an experimental battery of object free-access double-choice (FADC) and social interaction tests—using optogenetics, chemogenetics, calcium fiber photometry, multichannel recording electrophysiology, and multicolor mRNA in situ hybridization—we uncovered a cell type–specific cortico-subcortical brain circuit of the curiosity and novelty-seeking behavior. We found in mice that inhibitory neurons in the medial ZI (ZIm) are essential for the decision to investigate an object or a conspecific. These neurons receive excitatory input from the prelimbic cortex to signal the initiation of exploration. This signal is modulated in the ZIm by the level of investigatory motivation. Increased activity in the ZIm instigates deep investigative action by inhibiting the periaqueductal gray region. A subpopulation of inhibitory ZIm neurons expressing tachykinin 1 (TAC1) modulates the investigatory behavior.
Redressing imbalances in the kind of research that gets done and who gets credit for it
If we want good work to get done, we should credit those who do it. In science, researchers are credited predominantly via authorship on publications. But many contributions to modern research are not recognized with authorship, due in part to the high bar imposed by the authorship criteria of many journals. “Contributorship” is a more inclusive framework for indicating who did what in the work described by a paper, and many scientific journals have recently implemented versions of it. I will consider the motivation for and specifics of this change, describe the tenzing tool we created to facilitate it, and how we might want to support and shape the shift toward contributorship
Developing a mouse incentive delay task
Monetary incentive delay task (MID) is a well-validated human functional MRI task widely used in probing affective-motivational processes in psychiatric disorders. We are developing a mouse version of the MID task in order to facilitate translations of findings from the wealth of human imaging studies. This talk presents our task design and behavioural data from the ongoing work.
A reward-learning framework of knowledge acquisition: How we can integrate the concepts of curiosity, interest, and intrinsic-extrinsic rewards
Recent years have seen a considerable surge of research on interest-based engagement, examining how and why people are engaged in activities without relying on extrinsic rewards. However, the field of inquiry has been somewhat segregated into three different research traditions which have been developed relatively independently -- research on curiosity, interest, and trait curiosity/interest. The current talk sets out an integrative perspective; the reward-learning framework of knowledge acquisition. This conceptual framework takes on the basic premise of existing reward-learning models of information seeking: that knowledge acquisition serves as an inherent reward, which reinforces people’s information-seeking behavior through a reward-learning process. However, the framework reveals how the knowledge-acquisition process is sustained and boosted over a long period of time in real-life settings, allowing us to integrate the different research traditions within reward-learning models. The framework also characterizes the knowledge-acquisition process with four distinct features that are not present in the reward-learning process with extrinsic rewards -- (1) cumulativeness, (2) selectivity, (3) vulnerability, and (4) under-appreciation. The talk describes some evidence from our lab supporting these claims.
A Changing View of Vision: From Molecules to Behavior in Zebrafish
All sensory perception and every coordinated movement, as well as feelings, memories and motivation, arise from the bustling activity of many millions of interconnected cells in the brain. The ultimate function of this elaborate network is to generate behavior. We use zebrafish as our experimental model, employing a diverse array of molecular, genetic, optical, connectomic, behavioral and computational approaches. The goal of our research is to understand how neuronal circuits integrate sensory inputs and internal state and convert this information into behavioral responses.
Hypothalamic control of internal states underlying social behaviors in mice
Social interactions such as mating and fighting are driven by internal emotional states. How can we study internal states of an animal when it cannot tell us its subjective feelings? Especially when the meaning of the animal’s behavior is not clear to us, can we understand the underlying internal states of the animal? In this talk, I will introduce our recent work in which we used male mounting behavior in mice as an example to understand the underlying internal state of the animals. In many animal species, males exhibit mounting behavior toward females as part of the mating behavior repertoire. Interestingly, males also frequently show mounting behavior toward other males of the same species. It is not clear what the underlying motivation is - whether it is reproductive in nature or something distinct. Through detailed analysis of video and audio recordings during social interactions, we found that while male-directed and female-directed mounting behaviors are motorically similar, they can be distinguished by both the presence of ultrasonic vocalization during female-directed mounting (reproductive mounting) and the display of aggression following male-directed mounting (aggressive mounting). Using optogenetics, we further identified genetically defined neural populations in the medial preoptic area (MPOA) that mediate reproductive mounting and the ventrolateral ventromedial hypothalamus (VMHvl) that mediate aggressive mounting. In vivo microendocsopic imaging in MPOA and VMHvl revealed distinct neural ensembles that mainly encode either a reproductive or an aggressive state during which male or female directed mounting occurs. Together, these findings demonstrate that internal states are represented in the hypothalamus and that motorically similar behaviors exhibited under different contexts may reflect distinct internal states.
Exploring the neural landscape of imagination and abstract spaces
External cues imbued with significance can enhance the motivational state of an organism, trigger related memories and influence future planning and goal directed behavior. At the same time, internal thought and imaginings can moderate and counteract the impact of external motivational cues. The neural underpinnings of imagination have been largely opaque, due to the inherent inaccessibility of mental actions. The talk will describe studies utilizing imagination and tracking how its neural correlates bidirectionally interact with external motivational cues. Stimulus-response associative learning is only one form of memory organization. A more comprehensive and efficient organizational principal is the cognitive map. In the last part of the talk we will examine this concept in the case of abstract memories and social space. Social encounters provide opportunities to become intimate or estranged from others and to gain or lose power over them. The locations of others on the axes of power and affiliation can serve as reference points for our own position in the social space. Research is beginning to uncover the spatial-like neural representation of these social coordinates. We will discuss recent and growing evidence on utilizing the principals of the cognitive map across multiple domains, providing a systematic way of organizing memories to navigate life.
The structure of behavior entrained to long intervals
Interpretation of interval timing data generated from animal models is complicated by ostensible motivational effects which arise from the delay-to-reward imposed by interval timing tasks, as well as overlap between timed and non-timed responses. These factors become increasingly prevalent at longer intervals. To address these concerns, two adjustments to long interval timing tasks are proposed. First, subjects should be afforded with reinforced non-timing behaviors concurrent with timing. Second, subjects should initiate the onset of timed stimuli. Under these conditions, interference by extraneous behavior would be detected in the rate of concurrent non- timing behaviors, and changes in motivation would be detected in the rate at which timed stimuli are initiated. In a task with these characteristics, rats initiated a concurrent fixed-interval (FI) random-ratio (RR) schedule of reinforcement. This design facilitated response-initiated timing behavior, even at increasingly long delays. Pre-feeding manipulations revealed an effect on the number of initiated trials, but not on the timing peak function.
Fish Feelings: Emotional states in larval zebrafish
I’ll give an overview of internal - or motivational - states in larval zebrafish. Specifically we will focus on the role of the Oxytocin system in regulating the detection of, and behavioral responses to, conspecifics. The appeal here is that Oxytocin has likely conserved roles across all vertebrates, including humans, and that the larval zebrafish allows us to study some of the general principles across the brain but nonetheless at cellular resolution. This allows us to propose mechanistic models of emotional states.
Sleep and the gut
Sleep is generally associated with the brain but poor sleep impacts the entire body - many diseases are caused or exacerbated by sleep loss. Our work is uncovering ways in which sleep and the body interact. We found a special, two-way relationship between sleep and the gut: the gut is uniquely impacted by sleep loss, and it actively controls sleep quality. These findings could help us understand the origins of sleep as well as develop strategies to offset the negative consequences of inadequate sleep.
Harnessing Mindset in 21st Century Healthcare
Mindsets are core assumptions about the nature and workings of things in the world that orient us to a particular set of attributions, expectations, and goals. Our study of mindsets is, in part, inspired by research on the placebo effect, a robust demonstration of the ability of mindsets, conscious or subconscious, to elicit physiological changes in the body. This talk will explore the role of mindsets in three stages of chronic disease progression: genetic predisposition, behavioral prevention, and clinical treatment. I will discuss the mechanisms through which mindsets influence health as well as the myriad ways that mindsets can be more effectively leveraged to motivate healthy behaviors and improve 21st century healthcare.
Becoming Human: A Theory of Ontogeny
Humans are biologically adapted for cultural life in ways that other primates are not. Humans have unique motivations and cognitive skills for sharing emotions, experience, and collaborative actions (shared intentionality). These motivations and skills first emerge in human ontogeny at around one year of age, as infants begin to participate with other persons in various kinds of collaborative and joint attentional activities, including linguistic communication. Our nearest primate relatives understand important aspects of intentional action - especially in competitive situations - but they do not seem to have the motivations and cognitive skills necessary to engage in activities involving collaboration, shared intentionality, and, in general, things cultural.
Analogies, Games and the Learning of Mathematics
Research on analogical processing and reasoning has provided strong evidence that the use of adequate educational analogies has strong and positive effects on the learning of mathematics. In this talk I will show some experimental results suggesting that analogies based on spatial representations might be particularly effective to improve mathematics learning. Since fostering mathematics learning also involves addressing psychosocial factors such as the development of mathematical anxiety, providing social incentives to learn, and fostering engagement and motivation, I will argue that one area to explore with great potential to improve math learning is applying analogical research in the development of learning games aimed to improve math learning. Finally, I will show some early prototypes of an educational project devoted to developing games designed to foster the learning of early mathematics in kindergarten children.
Plasticity in hypothalamic circuits for oxytocin release
Mammalian babies are “sensory traps” for parents. Various sensory cues from the newborn are tremendously efficient in triggering parental responses in caregivers. We recently showed that core aspects of maternal behavior such as pup retrieval in response to infant vocalizations rely on active learning of auditory cues from pups facilitated by the neurohormone oxytocin (OT). Release of OT from the hypothalamus might thus help induce recognition of different infant cues but it is unknown what sensory stimuli can activate OT neurons. I performed unprecedented in vivo whole-cell and cell-attached recordings from optically-identified OT neurons in awake dams. I found that OT neurons, but not other hypothalamic cells, increased their firing rate after playback of pup distress vocalizations. Using anatomical tracing approaches and channelrhodopsin-assisted circuit mapping, I identified the projections and brain areas (including inferior colliculus, auditory cortex, and posterior intralaminar thalamus) relaying auditory information about social sounds to OT neurons. In hypothalamic brain slices, when optogenetically stimulating thalamic afferences to mimic high-frequency thalamic discharge, observed in vivo during pup calls playback, I found that thalamic activity led to long-term depression of synaptic inhibition in OT neurons. This was mediated by postsynaptic NMDARs-induced internalization of GABAARs. Therefore, persistent activation of OT neurons following pup calls in vivo is likely mediated by disinhibition. This gain modulation of OT neurons by infant cries, may be important for sustaining motivation. Using a genetically-encoded OT sensor, I demonstrated that pup calls were efficient in triggering OT release in downstream motivational areas. When thalamus projections to hypothalamus were inhibited with chemogenetics, dams exhibited longer latencies to retrieve crying pups, suggesting that the thalamus-hypothalamus noncanonical auditory pathway may be a specific circuit for the detection of social sounds, important for disinhibiting OT neurons, gating OT release in downstream brain areas, and speeding up maternal behavior.
Monkey Talk – what studies about nonhuman primate vocal communication reveal about the evolution of speech
The evolution of speech is considered to be one of the hardest problems in science. Studies of the communicative abilities of our closest living relatives, the nonhuman primates, aim to contribute to a better understanding of the emergence of this uniquely human capability. Following a brief introduction over the key building blocks that make up the human speech faculty, I will focus on the question of meaning in nonhuman primate vocalizations. While nonhuman primate calls may be highly context specific, thus giving rise to the notion of ‘referentiality’, comparisons across closely related species suggest that this specificity is evolved rather than learned. Yet, as in humans, the structure of calls varies with arousal and affective state, and there is some evidence for effects of sensory-motor integration in vocal production. Thus, the vocal production of nonhuman primates bears little resemblance to the symbolic and combinatorial features of human speech, while basic production mechanisms are shared. Listeners, in contrast, are able learning the meaning of new sounds. A recent study using artificial predator shows that this learning may be extremely rapid. Furthermore, listeners are able to integrate information from multiple sources to make adaptive decisions, which renders the vocal communication system as a whole relatively flexible and powerful. In conclusion, constraints at the side of vocal production, including limits in social cognition and motivation to share experiences, rather than constraints at the side of the recipient explain the differences in communicative abilities between humans and other animals.
The role of spatiotemporal waves in coordinating regional dopamine decision signals
The neurotransmitter dopamine is essential for normal reward learning and motivational arousal processes. Indeed these core functions are implicated in the major neurological and psychiatric dopamine disorders such as schizophrenia, substance abuse disorders/addiction and Parkinson's disease. Over the years, we have made significant strides in understanding the dopamine system across multiple levels of description, and I will focus on our recent advances in the computational description, and brain circuit mechanisms that facilitate the dual role of dopamine in learning and performance. I will specifically describe our recent work with imaging the activity of dopamine axons and measurements of dopamine release in mice performing various behavioural tasks. We discovered wave-like spatiotemporal activity of dopamine in the striatal region, and I will argue that this pattern of activation supports a critical computational operation; spatiotemporal credit assignment to regional striatal subexperts. Our findings provide a mechanistic description for vectorizing reward prediction error signals relayed by dopamine.
Neuroimaging in human drug addiction: an eye towards intervention development
Drug addiction is a chronically relapsing disorder characterized by compulsive drug use despite catastrophic personal consequences (e.g., loss of family, job) and even when the substance is no longer perceived as pleasurable. In this talk, I will present results of human neuroimaging studies, utilizing a multimodal approach (neuropsychology, functional magnetic resonance imaging, event-related potentials recordings), to explore the neurobiology underlying the core psychological impairments in drug addiction (impulsivity, drive/motivation, insight/awareness) as associated with its clinical symptomatology (intoxication, craving, bingeing, withdrawal). The focus of this talk is on understanding the role of the dopaminergic mesocorticolimbic circuit, and especially the prefrontal cortex, in higher-order executive dysfunction (e.g., disadvantageous decision-making such as trading a car for a couple of cocaine hits) in drug addicted individuals. The theoretical model that guides the presented research is called iRISA (Impaired Response Inhibition and Salience Attribution), postulating that abnormalities in the orbitofrontal cortex and anterior cingulate cortex, as related to dopaminergic dysfunction, contribute to the core clinical symptoms in drug addiction. Specifically, our multi-modality program of research is guided by the underlying working hypothesis that drug addicted individuals disproportionately attribute reward value to their drug of choice at the expense of other potentially but no-longer-rewarding stimuli, with a concomitant decrease in the ability to inhibit maladaptive drug use. In this talk I will also explore whether treatment (as usual) and 6-month abstinence enhance recovery in these brain-behavior compromises in treatment seeking cocaine addicted individuals. Promising neuroimaging studies, which combine pharmacological (i.e., oral methylphenidate, or RitalinTM) and salient cognitive tasks or functional connectivity during resting-state, will be discussed as examples for using neuroimaging for empirically guiding the development of effective neurorehabilitation strategies (encompassing cognitive reappraisal and transcranial direct current stimulation) in drug addiction.

Mammalian neonates are born immature. Thus mothers are equipped with innate motivation to nurture them. Moreover, in species that live in a family group, fathers and older siblings may also provide extensive care to the young. By studying those highly social species, including laboratory mice, common marmosets, and humans, we are trying to elucidate the neural mechanisms of parental care. Neuronal activity mapping and site-specific functional suppression in mice identified the central part of the medial preoptic area (cMPOA) as the hub of caregiving network for both mothers and fathers.Recent findings about the neural circuit and molecular signaling involved in caregiving motivation will be discussed.
Neural Circuit Mechanisms of Emotional and Social Processing
How does our brain rapidly determine if something is good or bad? How do we know our place within a social group? How do we know how to behave appropriately in dynamic environments with ever-changing conditions? The Tye Lab is interested in understanding how neural circuits important for driving positive and negative motivational valence (seeking pleasure or avoiding punishment) are anatomically, genetically and functionally arranged. We study the neural mechanisms that underlie a wide range of behaviours ranging from learned to innate, including social, feeding, reward-seeking and anxiety-related behaviours. We have also become interested in “social homeostasis” -- how our brains establish a preferred set-point for social contact, and how this maintains stability within a social group. How are these circuits interconnected with one another, and how are competing mechanisms orchestrated on a neural population level? We employ optogenetic, electrophysiological, electrochemical, pharmacological and imaging approaches to probe these circuits during behaviour.
