Neuronal development can be severely impacted under pathological conditions, altering the maturation of neuronal cells. Allan Herndon Dudley Syndrome (AHDS) is a rare X-linked syndrome caused by a mutation in the SLC16A2 gene, which encodes the specific transporter (monocarboxylate transported 8, MCT8) of thyroid T3 hormone, reducing its presence in the central nervous system and leading to severe neurological impairment. We investigated the neuronal maturation in an in vitro organoid model of AHDS, in which the pathological condition was resembled by removing the T3 hormone from the culture m

The C-terminal binding protein 1 (CtBP1) is a ubiquitously expressed, dual-function protein. In the nucleus, it nucleates corepressor complexes that control chromatin-modification and gene expression. In the cytoplasm, it regulates membrane trafficking processes by controlling the membrane fission reaction. Our previous study revealed an involvement of CtBP1 in the control of neural activity‐regulated genes such as BDNF, Arc and Fos in the mature brain. Recently, individuals with a missense mutation in CTBP1 have been reported to have intellectual disability and neurodevelopmental problems. Th

Nociceptors are a subset of sensory neurons that relay painful stimuli from the peripheral tissues. Multiple nociceptor subtypes exist and can respond to different thermal, mechanical, or chemical noxious stimuli. Recent findings with transcriptomics have identified gene expression patterns that classify these sub-types by different ion channels including NaV1.7, NaV1.8, NaV1.9, TRPV1, TRPA1, P2X3, HCN and a number of others that have been implicated in pain. In this study, we have identified a method to rapidly generate a novel TRPA1+ human induced pluripotent stem cell (hiPSC)-derived sensor

Serotonergic psychedelics are emerging as alternative therapies for treatment-resistant depression; however, their hallucinogenic effects may limit their widespread clinical use. Second-generation psychedelics, which lack hallucinogenic effects, are a promising alternative. 2-Bromo-LSD (2-Br-LSD/BETR-001) is an LSD derivative with 5-HT2A partial agonist activity that lacks hallucinogenic effects. Thus, 2-Br-LSD may represent a possible novel therapeutic for treating neuropsychiatric diseases. We assessed the effects of 2-Br-LSD on stress-coping behaviour in mice, using the forced swim test, an

Neuroinflammation is associated with many medical conditions that exhibit both CNS and behavioural changes. Understanding the biological mechanisms underpinning these inflammation-triggered changes may aid therapeutic discovery and further pathophysiological involvement in several neuropsychiatric conditions (e.g. depression and anxiety). Upon activation, toll-like receptor 7 (TLR7) triggers an interferon-mediated inflammatory response. In this study, we utilise the non-invasive, TLR7-activated Aldara Imiquimod mouse model to investigate the transcriptomic changes within the pro-inflammatory n
