Cryopreservation of Manufactured Cells by Encapsulation

Alptekin Aksan · University of Minnesota, Department of Mechanical Engineering

Cryopreservation constrains manufacturing and clinical delivery of immune cells such as NK and T cells. DMSO protects against freezing but becomes toxic at physiological temperatures; associated infusion morbidity can require washing or divided dosing that diminishes therapeutic effectiveness. This talk presents an alternative in which reversible, cytocompatible hydrogels encapsulate cells before freezing without DMSO. The speaker examines evidence that encapsulation cushions osmotic stress, suppresses kinetic and phase transitions, and reduces injury caused by extracellular ice growth and fus

Clinical grade production of large-scale neural progenitor cells (NPC) for Huntington’s disease treatment

Josep M. Canals, Marc Estarellas, Georgina Bombau et al. · FENS Forum 2024

Regenerative medicine aims to revolutionize medical practice by offering cures for severe diseases. This is achieved through the utilization of human pluripotent stem cell (hPSC) lines, which undergo controlled differentiation before being transplanted into patients. A significant challenge lies in developing safe, robust, and controlled differentiation protocols, requiring the adaptation of research-only reagents to good manufacturing practice (GMP) standards.Cell-based therapies for Huntington’s disease aspire to restore neuronal circuitry and function by replacing lost neurons. Here, we pre
