Intracerebral haemorrhage (ICH) is the second-most common form of stroke. Due to the post-ICH brain damage and limited therapeutic strategies, the disability rate among ICH survivors is high. DJ-1 is a protein known to play roles in cytoprotection. However, DJ-1 can be oxidised and lose its protective functions. Our study investigated the oxidation state of DJ-1 after ICH and the therapeutic potential of DJ-1 oxidation inhibitor, DJ-1-binding compound 23 (Comp-23), in ICH. We induced experimental ICH mice models by intra-striatal injection of collagenase. Immunoblot, qPCR measurement and immun

We aimed to model tau abnormalities in the early and advanced stages of Alzheimer’s disease and other tauopathies. By doing so, we wanted to evaluate molecules capable of preventing tau-mediated neurodegeneration. To achieve this, we established cultures of mouse cortical neurons in which hyperphosphorylated (but not aggregated) tau accumulates in a significant population of neurons following chronic exposure to redox-active iron from hemin (Hm), a breakdown product of hemoglobin. Using experimental conditions in which p-tau (AT8 immunosignal) accumulation occurs without concomitant neuronal l

Aims: Excessive oxidative stress is a major factor to damage retinal pigment epithelial (RPE) cells that promotes the development of retinal degeneration. We investigated the effects of 6dS, a pigment epithelium-derived factor (PEDF) derived factor on cultured RPE cells and retina of rats treated with sodium iodate (NaIO3). Methods: Annexin V/PI staining was used to determine ARPE-19 cell viability. FerroOrange and immunostaining of acrolein were used to detect intracellular labile ion (Fe2+)-dependent cell ferroptosis and lipid peroxidation reaction in plasma membrane. Immunoblotting and real

Almost all body functions undergo circadian variations synchronized by light/dark cycle-regulated melatonin secretion from the pineal gland. Circadian disruption reduces melatonin secretion and increases the risk of diabetes and its complications, including anxiety. We aimed to study the effects of reduced daily light exposure on diabetes-associated anxiety-like behavior and brain oxidative stress in streptozotocin-treated rats. Male Wistar rats were divided into the following groups: 1. control (C12/12), saline-treated group with light/dark cycle 12/12h; 2. saline-treated group on light/dark
