Aged microglia in Alzheimer’s disease display a senescent and pro-inflammatory profile associated with mitochondrial oxidative stress

Vicente Roca Agujetas, Jorge Moreno-Fernández, Cristina Núñez-Díaz et al. · FENS Forum 2024

Microglial phenotype during Alzheimer’s Disease (AD) continuum is under debate. At early stages, a protective phenotype with the typical DAM profile would participate in plaque compaction and Aβ degradation. Instead, human AD microglia (HAM) show an enhance aged/degenerative profile, supporting a microglial dysfunction as a central mechanism in AD aetiology. Recent findings from our lab described that peri-plaque microglia are highly reliant on oxygen levels and mitochondrial integrity. Nevertheless, mitochondrial implication in aged microglia during the disease progression remains unclear. Th

Regulation of repressor-element 1 silencing transcription factor (REST) by CaMKIV

Hanako Tsushima, Anna Rocchi, Corradi Beatrice et al. · FENS Forum 2024

…plasticity, neuroinflammation and senescence.Understanding the endogenous mechanisms that control the level of REST is critical, if the protective expression of REST in neurodegeneration or aging were to become the focus of therapeutic approaches. Calcium is a signaling molecule that regulates neuronal activities as well as cellular processes such as autophagy leading to senescen
